Abstract: We will review some of the popular methods for distance-based phylogeny reconstruction with a focus on the statistical theory underlying the methods. We present a characterization theorem of least squares methods that result in efficient estimators and discuss the implications for minimum evolution phylogeny and neighbor-joining.
